There are really two questions here:

(a) does the favicon have to be *named* "/favicon.ico"?; and
(b) does it have to be in Windows Icon format -- can it be in PNG?

A. No, it doesn't. But if you give it a different name, browsers won't
know where to look for it, so you'll have to provide a link to it in your
HTML files:

<link rel="shortcut icon"
href="/myfavicon.ico"
type="image/x-icon">

B. No, it doesn't. Some browsers support favicons in other image formats,
like PNG. However, Internet Explorer *only* supports Windows Icon format
for favicons.

In general, a favicon must be an icon. That's why it's call a favICON.

Taking a file that is not an icon and changing the extension (e.g., your
favicon.png) to ico will not work. In this case, the file is still
formatted as PNG.

On the other hand, none of my favicons are named "favicon.ico". As
Inkster points out, the LINK element can be used to indicate which of
several icons to use. I use seven different icons, depending on the
topic of the Web page.
