Clam AV is open source.  Download the source code and have a peek at
an example.

Clam on Linux is excellent.
ClamWin is awful.

I think ClamAV is a good start and a good suggestion for a question not easily
answered.

yeah, clamav isn't a good suggestion for a product to use (at least not
for desktop protection), but for the question the OP is asking (how to
make an anti-virus) it's probably the best place to *start*...
