 We just upgraded to win2k3 enterprise server. We configured one of the server
as File server and whenever the users access a file on the shared folder it
is taking too much time. It takes more time to save the file with small
chages. I played with permissions I can not seem to figure out.

Any thoughts?

 Have you set up a DNS server for the domain? You can't use your ISP's DNS
server.

Does your clients use the DNS server you set up for the AD domain ONLY?
Again you can't use your ISP's DNS servers.

Everything was working fine in Win2k. The issue started when we upgraded to
win2k3.

Yes we do have a domain controller with non-ISP Dns IP addresses.
