Slow performance

We configured one of the server as File server and whenever the users access a file on the shared folder it is taking too much time. It takes more time to save the file with small chages. I played with permissions I can not seem to figure out. Any thoughts? Does your clients use the DNS server you set up for the AD domain ONLY? The issue started when we upgraded to win2k3.
